- According to research article by PubMed Central, Patients with primary EGFR mutations in non-small cell lung cancer (NSCLC) are now best treated with epidermal growth factor tyrosine kinase inhibitors, or EGFR-TKIs. After about a year, the patients do, however, inevitably become resistant to the EGFR-TKIs. The insensitivity of EGFR-TKI in non-small cell lung cancer is frequently caused by the activation of other receptor-mediated signaling pathways in addition to the acquisition of the EGFR T790M mutation. A poor prognosis for many tumors is linked to upregulation of the Tyro3-Axl-MERTK (TAM) family member Mer receptor tyrosine kinase (MERTK). The signaling pathways that EGFR shares, phosphoinositide 3-kinase (PI3K)/Akt and mitogen-activated protein kinase (MAPK), are triggered when particular ligands, like Gas6 and PROS1, bind to MERTK.
