Digital PCR Market (dPCR) is the third generation of PCR which helps in enabling the absolute quantification through partitioning of reaction. Digital PCR (dPCR) is highly sensitive and accurate in molecular detection. Digital PCR has determined the application like rare mutation detection, trace DNA detection, and copy number variation. Digital PCR helps in enabling precise and highly sensitive quantification of nucleic acids. The technique is called as ‘Digital PCR’ because reaction will be ‘1’ or will not be ‘0’ which generates fluorescent signal. Number of target molecules present on array is divided by dilution factor for giving the target DNA concentration of original sample. Traditional PCR determines the amount of amplified PCR product at end of several PCR cycles. Digital PCR helps in measuring the number of target molecules by directly counting the positive fluorescence in compartments. Digital PCR takes about 3 to 5 days after drop off of sample. Digital PCR is different from conventional PCR because it counts the target molecules directly in digital format without relying on standards or endogenous controls. Digital PCR test is performed by diluting assay mixture and sample into thousands of small compartments. Dilution is performed so that target molecule concentration in each reaction can be assumed safely as zero or one. Thermal cycling is carried out at end point. Compartments that contains target molecule shows fluorescence while compartment which do not contains target molecules shows only background fluorescence. Reaction with target molecule is counted as one or called as PCR-positive reaction while the reaction without target molecule is counted as zero or called as PCR-negative reactions. After counting complete set of reactions number of target molecules in entire reaction volume is equal to the number of positive reactions which is counted. Thus, the absolute target concentration is then calculated by dividing the total number of target molecules by total volume. Independence on amplification efficiency & high repeatability of results - Digital PCR realize absolute quantification as it defines amplification results with ‘1’ and ‘0’ & therefore, quantifies results with counts, which makes possible absolute quantification. Digital PCR can retest the results of NGS – Digital PCR helps in calculating the expression of nucleic acids with end-point signals, and therefore its results rule out interference of unstable amplification & becomes highly repeatable with below 5% error rate, and lowers mispairing of target sequences. Digital PCR can be combined with NGS for quantifying and interpreting the data, and retest the results of NGS. In Digital PCR quality of sequencing data is guaranteed by including joint dimer, adaptors, long fusion junction, error like fragment, etc. However, Digital PCR suppresses former techniques in every aspects.

Droplet Digital PCR has wide applications like – in gynecological tumor, it has shown high con-cordance with traditional techniques and detect absolute copy number of HER2 in invasive breast cancer, second generation sequencing, far outdoing semi-quantitative PCR, and hybrid chip technologies. Digital PCR is transformed and widely used in obstetrics. Autosomal recessive disorder - Digital PCR can detect beta-thalassemia in case of maternal and paternal mutation, by quantifying maternal mutated DNA & fetal mutated DNA help in diagnosing the fetus inheriting pathogenic genes. For instance in 2012 (PMC), based on reformed relative mutation dosage and paternal haplotypes, Lam et al designed the probes for globulins & external SNP to pinpoint site of interest, digital PCR was performed for identifying genotype of fetus, and consequently two families of beta-thalassemia were screened out. Digital PCR has wide applications in clinical use for non-invasive prenatal testing, chromosomal aneuploidy detection, autosomal recessive disorder, gynecological tumor, HR-positive breast cancer, triple negative breast cancer, cervical cancer, etc. However, wide applications and various advantages of Digital PCR has fueled the target market growth.
