Whole Exome Sequencing (WES) works by selectively collecting and sequencing all of the genome's protein-coding regions (exons), which account for only 1-2% of the human genome yet include the majority of disease-causing mutations. Genomic DNA is initially fragmented, and exon-specific probes are employed to enrich these areas.
The enriched exonic DNA is then sequenced using high-throughput next-generation sequencing (NGS) platforms, and the resulting data is analyzed to identify genetic variants, such as single nucleotide variants (SNVs), insertions, deletions, and splice-site mutations, that may cause hereditary diseases or guide personalized treatments.
